We found thatthese cells express the TNF-α receptor TNFR1. NF-κB was present, and wasactivated by exposure of cells to H O (250µM caused a 2.2-fold increase within2 25min), TNF-α (10ng/ml; 1.8-fold in 30min) or IL-1β (10ng/ml; 2.6-fold in 60min) in atime- and concentration dependent manner. The NF-κB subunits p65 and p50were involved in the rapid activation by TNF-α, and transfection of cells withantisense oligonucleotides against p65 subunit mRNA efficiently blocked thisresponse.
